| Using Rehmannia glutinosa seeds as test materials, standard germination test methods were employed to study the effects of different light conditions, temperatures, and germination substrates on the germination of R. glutinosa seeds, with the aim of exploring germination characteristics, determining optimal germination conditions, and evaluating the germination vigor of half-sibling seeds produced by different varieties.The results showed that light, temperature and germination substrate had significant effects on seed germination, and their interactions were also significant.The highest average membership function value for germination rate and germination index was 0.95 under the condition of light × 25°C × top-of-paper. Significant differences in seed vigor indices were observed among the five tested cultivars, with heritable values exceeding 72% for each index. Membership function analysis ranked the seed vigor of the tested varieties as follows: Hongshuwang > Sankuai > Shizitou > Xinfeng > Jinjiu. Based on seed vigor, Hongshuwang was classified as high vigor, Sankuai and Shizitou as medium vigor, and Xinfeng and Jinjiu as low vigor types. In conclusion, light×25°C×top of paper is the optimal condition for the germination of R. glutinosa seeds. The seed vigor of R. glutinosa can be highly hereditary, and the selection of high-vigor seed varieties through seed vigor evaluation can effectively promote the breeding of R. Glutinosa. |
