基于生物碱类成分结合多元统计分析评价甘肃不同产地栽培半夏药材的质量特征

      利用多元统计方法分析甘肃不同产地栽培半夏中9种生物碱的含量特征,发掘甘肃不同产地半夏中生物碱类化合物的含量与产地之间的关系。采用高效液相色谱法对来自甘肃4个主产区的39份半夏样品中的葫芦巴碱、烟碱、次黄嘌呤核苷酸、尿苷、次黄嘌呤核苷鸟苷、腺苷、胸苷、盐酸麻黄碱9种生物碱成分进行含量测定,结合非参数检验、相关性分析、主成分分析、聚类分析、判别分析等对含量与产地之间的关系进行分析评价。结果表明,39批半夏样品中除次黄嘌呤核苷酸的含量呈正态分布外,其余8种均呈偏态分布。Kruskal Wallis检验结果表明半夏不同产地半夏中9种生物碱成分含量存在显著差异( P<0.05);秦州区和西和县的半夏总体生物碱含量较高;相关性分析表明各生物碱成分之间存在复杂的关联性;聚类分析结果与Kruskal Wallis检验及主成分分析结果基本一致;OLPS-DA分析结果与主成分分析结果相互印证了烟碱、葫芦巴碱、尿苷、次黄嘌呤核苷酸是半夏的主要生物碱特征成分。因此甘肃不同产地半夏的生物碱质量特征存在差异,本研究为甘肃半夏的产地鉴别、质量控制及产业高质量发展提供依据。

      Multivariate statistical method was used to analyze the content characteristics of nine alkaloids in Pinellia ternata cultivated in different regions of Gansu province,and the relationship between alkaloid content and geographical origin was explored. High-performance liquid chromatography (HPLC) was used to determin the concentrations of nine alkaloids,including trigonelline,nicotine,5’inosinic acid,uridine,adenosine,inosine,vernine,thymidine and ephedrine in 39 pinellia samples from four main producing areas in Gansu Province. Non-parametric test,correlation analysis,principal component analysis,cluster analysis and discriminant analysis were used to analyze and evaluate the relationship between alkaloid content and geographical origin. The results showed that,except for the content of 5’Inosinic acid,which followed a normal distribution,the other eight alkaloids were skewed. The contents of nicotine,trigonelline,uridine,and 5’inosinic acid were relatively higher than those of the other alkaloids. .The Kruskal Wallis test showed significant differences in alkaloid content among Pinellia ternata samples from different producing areas (P<0.05). The alkaloid content was higher in Qinzhou District and Xihe County. Correlation analysis revealed complex interrelationships among alkaloid components. Using an eigenvalue greater than 1 as the criterion,three principal components were extracted through principal component analysis,with a cumulative variance contribution rate of 75.2%. The results of cluster analysis were largely consistent with those of the Kruskal-Wallis test and principal component analysis. The results of OPLS-DA analysis and principal component analysis mutually confirmed that nicotine,trigonelline,uridine,and 5′-inosinic acid were the main characteristic alkaloid components of Pinellia ternata. In conclusion,the alkaloid quality characteristics of pinellia pinellia from different producing areas in Gansu province exhibit significant differences. This study provides scientific basis for identification,quality control and high quality development of pinellia from Gansu Province.
